Cryo methods associated with electron microscopy vary from techniques that lead directly to the cryo electron microscope to those that bring the sample back to room temperature for imaging. The thing they have in common is the need to obtain a sample that is embedded in amorphous ice, a vitreous sample. A vitrified sample is in an aqueous state and therefore close to the native state. In this presentation, Leica experts will discuss Leica Microsystems solutions to achieve vitrification and to process it further while maintaining the native state. The evolution of Correlative Light and Electron Microscopy leading to the Leica Cryo CLEM, the cryo light microscope where a sample is imaged in the vitrified state and then transferred into a Cryo TEM for Correlative Light and Electron Microscopy or Cryo Tomography will be highlighted.

Correlative light and electron microscopy is an imaging technique that enables identification and targeting of fluorescently tagged structures with subsequent imaging at near-to-nanometer resolution.… The most significant and powerful characteristic for cell biology research is an ability to study the same cell using two different microscopy platforms: fluorescence light microscopy (FLM) that provide dynamic information on specific biomolecules and Electron Microscopy (EM) that provides detailed, high-resolution information on cellular ultra-struture and protein structure both under cryo-condition. During this talk, we will discuss the CryoCLEM principles and workflows.
